ELN - Analytical Technique

Chromatography

Purpose

Column chromatography is a technique applied to an experiment to purify compounds according

to their polar magnitude. To achieve successful column chromatography it is important to avoid

contamination.

Notes

- Ensure the there is liquid in the column at all times

- Do not allow the liquid to reach below the silica gel line, continue to refill

- Do not lose any of the liquid product dripping out

Protocol

Conditioning of the column

● Fill the top of the mini-column with 10%-90% ethyl acetate/hexane solvent.

● Obtain a pipet bulb and apply it to the top of the column, squeeze the bulb to force

solvent through the gel portion of the column

● When removing the pipet, maintain pressure and pull the bulb off to avoid inverse

pressure flow

● Do not allow the solvent level to drop below the silica line at the top
