List the formulation factors that can affect drug absorption by the buccal and sublingua routes.

Sublingual - best onset of action biggest advantage Fast-dissolving tablet ○ Protect from heat, light and moisture

Products can contain Lactose monohydrate, Glyceryl monostearate, pregelatinized starch, calcium steerage, NF powder, silicon dioxide and colloidal.

Excipients more hydrophilic or hydrophobic? Hydrophilic, because under the tongue you need to have fast dissolution

Spray ○ NitroMist

Products can contain Capellini/capric diglycerol succinct, pappermint oil, menthol, and n- butane

Buccal - for proteins, we need contact time Buccal tables Fentora Releases CO2(effervescent) to help with transient delivery(permeation enhancement)

Transient decrease in pH to optimize dissolution & increase in pH for membrane permeation

Buccal patch ○ Onsolis Buccal patch

Onsolis Buccal patch § For more drug, surface area is increased not mg dose. Mucoadhesive is critical for enhanced contact time with the Buccal epithelia

BOTH: no first pass
